Abstarct According to a highly
conserved 8 amino acid sequence of L proteins in Rhabdoviridae, a 24-mer
degenerate oligonucleotide primer was designed to amplify the 5′ terminus of
the genomic RNA of wheat rosette stunt virus (WRSV), a plant rhabdovirus, by
anchored PCR.The complete nucleotide sequence of the 5′ trailer in WRSV genomic
RNA was determined by deoxynucleotide sequencing of cloned cDNA derived from
the anchored PCR products.
